EPCO-09. TRANSCRIPTOMIC LANDSCAPE OF LOWER GRADE GLIOMA BASED ON AGE-RELATED NON-SILENT SOMATIC MUTATIONS

چکیده

Abstract Glioma accounts for 80% of all malignant brain tumours and is the most common adult primary tumour. Age an important factor affecting development cancer, as somatic mutations accumulate with age. Here, we aimed to analyse significance age-dependent non-silent in glioma prognosis. Histological tumour grade depends on age at diagnosis patients IDH1, TP53, ATRX, EGFR mutations. wild-type IDH1 increased increase grade, while or mutation remained constant. However, was higher than that mutation. The hierarchical clustering dominantly separated by Furthermore, were TP53 ATRX double its counterpart. lower TP53. Patients showed poorer prognosis those wild type genotype. Unlike mutant, upregulation expression epithelial mesenchymal transition associated genes.

Age-related somatic mutations in the cancer genome

Aging is associated with an increased risk of cancer, possibly in part because of an age-related increase in mutations in normal tissues. Due to their extremely low abundance, somatic mutations in normal tissues frequently escape detection. Tumors, as clonal expansions of single cells, can provide information about the somatic mutations present in these cells prior to tumorigenesis. Here, we us...

Molecular Subtypes of Glioblastoma Are Relevant to Lower Grade Glioma

BACKGROUND Gliomas are the most common primary malignant brain tumors in adults with great heterogeneity in histopathology and clinical course. The intent was to evaluate the relevance of known glioblastoma (GBM) expression and methylation based subtypes to grade II and III gliomas (ie. lower grade gliomas). METHODS Gene expression array, single nucleotide polymorphism (SNP) array and clinica...

Protein Domain-Level Landscape of Cancer-Type-Specific Somatic Mutations

Identifying driver mutations and their functional consequences is critical to our understanding of cancer. Towards this goal, and because domains are the functional units of a protein, we explored the protein domain-level landscape of cancer-type-specific somatic mutations. Specifically, we systematically examined tumor genomes from 21 cancer types to identify domains with high mutational densi...

Landscape of somatic mutations in sporadic GH-secreting pituitary adenomas.

CONTEXT Alterations in the cAMP signaling pathway are common in hormonally active endocrine tumors. Somatic mutations at GNAS are causative in 30-40% of GH-secreting adenomas. Recently, mutations affecting the USP8 and PRKACA gene have been reported in ACTH-secreting pituitary adenomas and cortisol-secreting adrenocortical adenomas respectively. However, the pathogenesis of many GH-secreting ad...

Somatic intronic microsatellite loci differentiate glioblastoma from lower-grade gliomas

Genomic studies of glioma sub-types have amassed new disease specific mutations, yet these only partially explain how mutations are linked to predisposition or progression. We hypothesized that microsatellite variation could expand the understanding of glioma etiology. Furthermore, germline markers for gliomas are typically undetectable; therefore we also hypothesize that the predictability of ...
